it depends if you can fit your hand between the gap or not. it also depends on the kind of rim you have, some you can stick your hands between the spokes and some you cant. Its hellava lot easier trying to adjust it with the rim off though
I would just spend an extra minute or 2o to take the tire off, in the long run it'll be a lot quicker w/o the tire on rather than working around the tire to adjust it. For me it's easier anyways.
